Focus on HER3 in ovarian cancer
How common is HER3 overexpression in ovarian cancer?
To date, few studies have measured the prevalence of HER3 expression in ovarian cancer.
| Source | N | Definition of HER3 overexpression | Percentage of tumors with HER3 overexpression |
|---|---|---|---|
| Tanner et al, J Clin Oncol, 20062 | 116 | Rajkumar score >8 (note: a Rajkumar score of 8 was the median value in this study) | 53% |
| Rajkumar et al, Clin Mol Pathol, 199611 | 98 | Higher HER3 expression than in normal ovarian epithelium by IHC | 16% |
Additional studies are needed to determine the prevalence of HER3 overexpression in ovarian cancer.
HER3 interacts with other HER family receptors in ovarian cancer
Preclinical research suggests the presence of HER3 interactions specific to ovarian cancer. These include4:
- Prolonged activation of the MAPK survival pathway in response to activation of both HER3 and HER4
- Activation by the ligand heregulin independent of HER2 and HER4
The clinical significance of these findings is unclear.
